Forskolin directly, rapidly, and reversibly activates adenylate cyclase, leading to a significant increase in intracellular cAMP levels. This rise in cAMP enhances body fat utilization, increases basal metabolic rate, and regulates the body’s thermogenic response to food. Consequently, forskolin supplements may promote fat loss without reducing muscle mass.

A topical cosmetic slimming product has been developed, combining forskolin, tetrahydroxypropyl ethylenediamine, caffeine, carnitine, and retinol. A clinical trial was conducted on five selected body areas among 78 women. Coleus forskohlii is a perennial herb from a large genus of mints. It is an important traditional Ayurvedic herb that has been used in Indian medicine for centuries. Coleus is the source of a unique compound known as forskolin.

The topical application of forskolin to a murine model with humanized skin and a mutant MC1R has resulted in significant melanization. This study suggests that forskolin is a potential candidate for clinical use in sunless tanning products. Additionally, it may serve as a topical small-molecule treatment for pigmentation that can protect against UV-induced cutaneous DNA damage.
